The SPR assay provides a method for determining the affinity and binding kinetics of a ligand to its receptor and measures the real-time binding association and dissociation rates. In utilizing this strategy, one of the binding partners (e.g., receptor protein) is immobilized on a biosensor surface. In contrast, the other partner, e.g., drug ligand, is continuously flown across the biosensor surface and binds to the immobilized receptor. Binding is measured as a change in resonance units (RUs) on the biosensor surface, and measuring the upregulation of bounded ligands over time at a given ligand concentration reveals the ligand association rate (ka or Kon). Ceasing the flow of the drug ligand and switching buffer alone allows the ligand to wash off from the receptor. Meanwhile, measuring the downregulation of bounded ligands over time reveals the ligand dissociation rate (kd or Koff). The affinity of the ligand for the receptor (the equilibrium dissociation constant, KD) is calculated based on these kinetic association rates and dissociation rates (kd/ka) by using multiple ligand concentrations.

The Biacore platform can provide detailed information on bispecific binding interactions and kinetic studies without the use of marker molecules. While the response curve of the system is displayed during the course of analysis, the change of resonance is susceptible to the changes at the surface of the sensor chip. The sensor chip is a vital part of the Biacore platform, which determines the system's sensitivity. Molecules can be immobilized on the surface of the sensor chip through different approaches, such as high-affinity capture (non-covalent interaction), covalent chemical interaction, and hydrophobic adsorption. Hence, this sensor chip has various functions to meet the needs and requirements of different projects.

Strategies for target analyte capturing rely on the interaction between the analyte and the ligand on the sensor surface. Some common capturing approaches are streptavidin/avidin-biotin interaction, antibody-antigen interaction, and tagged recombinant proteins based capturing.
